چکیده

The vision of using electron tomography (ET) for studying the 3D organization of biological materials – from molecules to cells – emerged in the late 1960s. However it was only in the 1990s that this vision began to materialize. Cryogenic sample preparation techniques replaced the artifact-prone techniques of the past, and automated acquisition schemes made possible the recording of tomographic data sets at subcritical electron dose. The development of advanced image processing methods allowed to extract a maximum of information from noisy tomograms. Today, at resolutions of 2-4 nm, cryo-ET provides unprecedented insights into the molecular organization of cellular landscapes and helps to bridge the gap between molecular and cellular structural studies performed in situ. Here we review milestones in the development of ET which made this

Artifact Correction for Zernike Phase-Plate Cryo-Electron Tomography

In cryo-electron microscopy (cryo-EM), vitreously frozen biological specimens can tolerate only a limited electron dose for imaging. Therefore, cryo-EM micrographs display weak contrast and a poor signal-to-noise ratio. This limits data interpretability and prevents efficient application of cryo-EM to a wider range of biological problems. Continued efforts have been invested in imaging methods ...

Vision in the Small: Reconstructing the Structure of Protein Macromolecules from Cryo-Electron Micrographs

Single particle reconstruction using Cryo-Electron Microscopy (cryo-EM) is an emerging technique in structural biology for estimating the 3-D structure (density) of protein macromolecules. Unlike tomography where a large number of images of a specimen can be acquired, the number of images of an individual particle is limited because of radiation damage. Instead, the specimen consists of identic...

Correlative cryo-fluorescence light microscopy and cryo-electron tomography of Streptomyces.

Light microscopy and electron microscopy are complementary techniques that in a correlative approach enable identification and targeting of fluorescently labeled structures in situ for three-dimensional imaging at nanometer resolution. Correlative imaging allows electron microscopic images to be positioned in a broader temporal and spatial context. We employed cryo-correlative light and electro...

Zernike Phase Contrast Cryo-Electron Tomography of Bacteria and Viruses

Cryo-electron tomography (cryo-ET) is currently the only method available through which pleomorphic biological specimens are imaged to produce three-dimensional (3D) structures at ~2 to 10 nm resolution [1]. There remain challenges associated with imaging intact bacteria and pleomorphic viruses by cryo-ET. In particular, investigators cope with the low contrast of cryo-preserved, unstained spec...

Mind the Gap: How Some Viruses Infect Their Hosts

Cryo-electron microscopy (Cryo-EM) and cryo-electron tomography (Cryo-ET) provide structural insights into complex biological processes. The podoviridae are dsDNA containing phage with short, non-contractile tails which nevertheless translocate their DNA into the cytoplasm of their host cells. Liu et al. [1] used a combination of cryo-EM and cryo-ET to study the structural changes accompanying ...
